People arriving on Island will see different rules to those which were imposed by the Manx Government last month.
Incoming residents will be tested on Day One, Day 6 or 7 and Day 13. Chief Minister Howard Quayle confirmed each test will be charged at £50.
Those who do not wish to be tested will be required to isolate for 21 days.
The changes are in line with the Level 5 border restrictions and will come into force from Wednesday night into Thursday morning.
